摘    要:目的观察白藜芦醇对人食管癌Eca109细胞增殖影响及凋亡诱导作用。方法噻唑蓝法测定细胞生长抑制率,倒置显微镜、透射电镜和HE染色法观察细胞的形态变化,流式细胞术用于检测白藜芦醇培养Eca109细胞前后的细胞凋亡情况。结果白藜芦醇对Eca109细胞生长有抑制作用,抑制率达76.42%,并呈剂量、时间效应关系。倒置显微镜下可见细胞生长明显被抑制,胞浆内颗粒增多增粗,部分细胞变圆悬浮在培养基中;HE染色和透射电镜观察发现白藜芦醇能诱导Eca109细胞呈现典型的细胞凋亡特征,如细胞体积变小,染色质浓缩等。流式细胞术检测结果显示,40 mg/L白藜芦醇培养Eca109细胞72 h后细胞凋亡率达19.52%,在DNA直方图上有明显的亚二倍体凋亡峰。结论白藜芦醇能明显抑制Eca109细胞增殖,并进一步诱导食管癌细胞凋亡。

Abstract:Objective To explore the inductive effects of resveratrol on the growth and apoptosis of human esophageal cancer Eca109 cells.Methods After different concentrations of resveratrol treated Eca109 cells for 24,48 and 72 hours,3-(4,5-dimethyl-2thiahiazoy)-3,5dy-phenyl-tetrazolium bromide(MTT) assay was used to determine the cell growth inhibitory rate.The changes of morphology was observed by inverted phase contrast microscope,transmission electron microscope and HE staining method.Flow cytometry were used to detect the apoptosis of Eca109 cells before and after resveratrol treatment.Results Resveratrol is able to inhibit the growth of Eca109 cells in a dose-dependent and time-dependent manner,and the inhibit rate arrived at 76.42%.The growth of durg-treated cells were inhibited and cell granulations were incereased and thicken,some cells became round ans suspension in the medium observed by invertedphase contrast microscope.transimisssion electron microscpoe and HE staining manner showed that resveratrol induced Eca109 cells to undergo apoptosis with typically apoptotic characteristics,including morphological changes of nuclear chromatin condensation and the bulk of nuclear lessening.Flow cytometry assay showed that after the treatment of Eca109 cells with resveratrol(50 mg/L) for 72 hours,the apoptotic rate reached at 19.52% and hypoploid apoptotic peak presented to the DNA picture.Conclusion Resveratrol could inhibit the proliferation of Eca109 cells evidently and induce the cells to apoptosis.
