消痈溃得康对胃溃疡模型大鼠生长因子和凋亡相关因子的影响

摘    要:目的:探讨中药消痈溃得康对乙酸所诱导的大鼠胃溃疡模型治疗作用的分子机制.方法:将40只大鼠随机分为正常组、假手术组、模型组、消痈溃得康组和奥美拉唑组,每组8只.采取胃黏膜局部投与乙酸方法制备胃溃疡模型,并于造模后第2天开始给药,消痈溃得康组按照4 g·kg-1剂量灌胃给予消痈溃得康颗粒,奥美拉唑组按照1.25 g·kg-1剂量给予奥美拉唑.假手术组、模型组给予纯水,每日一次,共灌胃10 d.10d后取血清及胃组织进行检测.采用酶联免疫吸附(ELISA)方法检测血清中血管内皮生长因子(VEGF)、表皮生长因子(EGF)及与凋亡相关的因子Bcl-2、C-myc的含量.取溃疡局部胃组织,Trozol法提取RNA,反转录为cDNA后采用实时荧光定量PCR法检测胃组织中EGFmRNA表达.结果:假手术组与正常组相比大鼠血清中EGF,VEGF,Bcl-2,C-myc含量没有显著差异;模型组中EGF,Bcl-2含量低于正常组(P<0.05),VEGF,C-myc的含量高于正常组(P<0.05);消痈溃得康组、奥美拉唑组大鼠血清中EGF,VEGF,Bcl-2含量高于模型组(P<0.05);消痈溃得康组、奥美拉唑组大鼠血清中C-myc含量低于模型组(P<0.05).荧光定量PCR检测EGFmRNA表达结果显示:假手术组与正常组相比没有差异,中药消痈溃得康治疗组大鼠胃组织EGF mRNA表达水平明显高于模型组(P<0.05).结论:中药消痈溃得康可以提高大鼠血清中VEGF,EGF等生长因子的表达,促进胃黏膜上皮及血管内皮细胞的生长,促进愈合,提高Bcl-2水平,抑制凋亡.同时降低C-myc水平,减少了癌变发生几率.

Abstract:Objective:To study the molecular mechanism of the treatment in which Xiaoyong Kuidekang would be applied to a model of gastric ulceration in rats. Method: Rats were divided into 5 groups randomly:normal group,sham group,ulcer model group, Xiaoyong Kuidekang group and Omeprazole group.Eight rats in each group.Acetic acid was applied to a certain part of the gastric mucosa to establish the ulcer model.The Xiaoyong Kuidekang treatment group had been administrated with drugs 4 g·kg-1and the Omeprazole group had been administrated with drugs 1.25 g·kg-1 since the second day after the operation.after 10 days The stomach and the serum was taken.The healing process of gastric ulcers was observed and the areas was measured.The content of EGF,VEGF,Bcl-2,and C-myc was measured by ELISA method.Total RNAs were extracted from the ulcer part of the gastric tissue by Trizol method,then the RNAs were transcribed reversely into cDNAs.The relative expression levels of EGF mRNA was measured by the QPCR assay. Result: There was no difference between the normal group and the sham operation group on levels of EGF,VEGF,C-myc and Bcl-2.the concentration of EGF,Bcl-2 in model group were lower than those in the normal group(P<0.05)and The concentration of VEGF,C-myc in model group were higher than those in the normal(P<0.05).The concentration of EGF,VEGF or Bcl-2 in the Xiaoyong Kuidekang group and in the Omeprazole group were much higher than those in the ulcer model group(P<0.05).The concentrations of C-myc in the Xiaoyong Kuidekang group and in the Omeprazole group were much lower than those in the ulcer model group(P<0.05).The mered result of the the rat EGFmRNA expression through the QPCR method is revealed.The expression of EGFmRNA in the Xiaoyong Kuidekang group were much higher than that in the ulcer model group(P<0.05). Conclusion: Xiaoyong Kuidekang can raise the expression of VEGF, EGF in rat serum, promote the growth of gastric epithelial and endothelial cell, promote healing, raise the level of Bcl-2, inhibit apoptosis.At the same time, reduce the level of C-myc, reducing the probability of occurrence of carcinogenesis.
