口腔鳞癌引流区淋巴结中树突状细胞特征性表型的免疫组化分析

摘    要:目的:探讨OSCC的DLN中DC的功能状态。方法:免疫组化标记OSCC的DLN和扁桃体组织中DC的CD1 a,CD83分子。统计CD1 a DC和CD83 DC的浸润程度,以及CD83 DC/CD1 a DC的比值。结果:CD1 a DC在OSCC的DLN中的浸润程度显著高于扁桃体组织(P<0.01),在鳞癌转移的淋巴结中显著低于未转移的淋巴结(P<0.01)。CD83 DC浸润程度在OSCC的DLN中明显低于扁桃体(P<0.01),但鳞癌转移的淋巴结与未转移者无显著差异(P>0.05)。OSCC的DLN中CD83 DC/CD1 a DC的比值显著低于扁桃体组织(P<0.05)。结论:OS-CC的DLN中,DC以不成熟DC为主,难以实现其激活T淋巴细胞反应的能力。

Abstract:Objective: To evaluate the functional state of dendritic cell(DC) in the draining lymph nodes of patients with oral squamous cell carcinoma.Methods: Immunohistochemistry was used to detect CD1a and CD83 molecules expressed by DC in paraffin embedded tissues from 34-case draining lymph nodes of OSCC and 10-case tonsils.DC counts,representing the density of DC infiltrating in the tissues,was performed under high-power field.The densities of DC expressing CD1a and CD83 molecules,and the ratio of CD83~ DC/CD1a~ DC were compared between two groups.Results: There were CD1a~ DCs in all cases.The density of CD1a~ DCs in the group of draining lymph node was significantly higher than the one of tonsil(P<0.01),but it was lower in draining lymph nodes with metastasis than without metastasis.The density of CD83~ DC was significantly lower in draining nodes than tonsils(P<0.01),but there was no significant difference between the nodes with metastasis and without metastasis(P>0.05).The ratio of CD83~ DC/CD1a~ DC was significantly higher in draining lymph nodes(P<0.05) than tonsils.Conclusion: Immature DC was predominant in the draining lymph nodes of OSCC,and the functional mature of DC was blocked so that DC could have difficulty in activating T lymph cells.
