霍乱弧菌非O1群菌株的毒力基因检测和染色体酶切图谱分析

摘    要:非O1群霍乱弧菌(不包括O139菌)是腹泻病原菌,由于它们分布较广,因此对人们的健康具有较大的威协;应用核酸技术对98株非O1菌的主要毒力基因进行检测,并从DNA和基因水平对5株非O1菌的分子特征进行研究。结果:所选菌株的染色体酶切图谱与O1群霍乱弧菌相比有很大差异,尤其与O1群流行株的差异更大,非O1菌相互之间的差异也很大;对98株非O1菌进行ctx、zot、ace、RS1基因检测,结果有2株为上述基因阳性,仅为2%左右。结果显示:非O1菌群体遗传结构复杂,变异程度高;其与O1菌,尤其是流行株(包括O139菌)的遗传关系较远;结果也说明大多数非O1菌是不致病的

Abstract:Vibrio cholerae (VC) non O1 (excluding VC O139) can be isolated from the environmental water almost through out the world. Part of them are able to cause human diarrhea. Furthermore some of the researchers considered that VC non O1 are the sources of the new type of Vibrio cholerae. So VC non O1 are the potential pathogens of the humans. In this paper 98 isolates of VC non O1 were detected for ctx, zot, ace and RS1 with colony hybridization and 5 isolates of them were studied with pulsed field gel electrophoresis. The results showed that there were significant differences between the chromosomal DNA restriction patterns of VC non O1 and VC O1, especially the epidemigenic strains of VC O1; By the colony hybridization with the gene probes of ctx, zot, ace and RS1, only two of ninety eight isolates of VC non O1 were positive. These results confirmed that only a very little part of VC non O1 which possess the identical molecular characteristics and virulence genes are pathogenic to humans.
