| 摘 要： | 目的:研究TNF-α对O-2A祖细胞、原少突胶质细胞、少突胶质细胞(oligodendrocytes,OL)这三个不同发育阶段少突胶质谱系细胞的易损性。方法:采用TNF-α分别刺激这三个不同发育阶段的细胞,MTT法检测存活率,免疫细胞化学检测活化的caspase-3。结果:O-2A祖细胞与OL相比存活率明显降低,原少突胶质细胞与OL相比存活率也降低;50ng/mlTNF-α刺激少突胶质谱系细胞48h以内,O-2A祖细胞的存活率与原少突胶质细胞、OL的存活率相比差异无统计学意义,而原少突胶质细胞的存活率与OL存活率相比较明显降低;caspase-3免疫细胞化学染色显示在这些细胞的胞浆和胞核有大量阳性信号。结论:TNF-α降低少突胶质谱系细胞的存活率具有细胞成熟程度的依赖性;caspase-3参与TNF-α诱导的3个不同发育阶段的OL的凋亡。
