苦丁茶熊果酸对鼻咽癌细胞增殖的抑制作用研究

摘    要:目的 探讨苦丁茶熊果酸对鼻咽癌细胞的抑制作用,为苦丁茶的深加工提供研究依据.方法 从苦丁茶老叶中提取熊果酸并纯化与鉴定,用5,10,20,40,80,100μmol·L-6个梯度浓度的熊果酸,持续作用于在培养的鼻咽癌细胞24,48,72,96,120 h后,用四甲基偶氮唑蓝比色法检测熊果酸对鼻咽癌细胞的抑制作用情况,流式细胞术分析其细胞周期和凋亡率.结果 不同浓度的苦丁茶熊果酸在不同时间内对鼻咽癌细胞均具有较强的抑制作用,并随浓度的升高及作用时间的延长而作用增强;流式细胞术结果显示,苦丁茶熊果酸阻滞鼻咽癌细胞于G0/G1期;40 μmol/L熊果酸作用48 h后抑制效果逐渐明显.结论 从苦丁茶提取的熊果酸对鼻咽癌细胞有较强的抑制作用,能阻滞癌细胞于G0/G1期,并诱导肿瘤细胞凋亡,苦丁茶极具开发前景.

Abstract:Objective To observe the toxicity effect,apoptosis induction and cell cycle inhibition of ursolic acid on human nasopharyngeal carcinoma cell,and to explore the posibility of Kuding tea as a new tumor chemopreventive drug.
