玄参活性部位对冠状动脉结扎致心室重构大鼠心肌纤维化的影响

摘    要:目的:探讨玄参活性部位对冠状动脉结扎所致心室重构大鼠心肌纤维化的影响及其作用机制。方法:冠状动脉结扎法制备大鼠心室重构模型。14周药物干预后,测定大鼠左心室及全心肥厚指数,病理切片HE染色观察心肌细胞横断面面积,天狼星红染色观察心肌间质及血管周围胶原沉积情况,并分析Ⅰ型、Ⅲ型胶原的分布和含量,荧光RT-PCR测定心肌组织Ⅲ型胶原基因表达水平。结果:玄参活性部位能降低心肌肥厚指数,减小左心室心肌细胞的横断面面积,改善心肌纤维化和胶原沉积,显著降低Ⅲ型胶原mRNA表达水平。结论:玄参活性部位对心肌细胞和间质胶原沉积两方面都有显著的抑制作用,抗心室重构的作用机制可能与抑制Ⅲ型胶原mRNA表达有关。

Abstract:Objective:To explore the impact of active sites of Radix Scrophulariae on myocardial fibrosis in ventricle-reconstructed rats induced by coronary ligation and its mechanism.Methods:The ventricle-reconstructed rat model was prepared with the method of coronary artery ligation.After 14 weeks of drug intervention,the hypertrophy index of left ventricle and the whole heart was determined;cross-sectional area of the myocardial cells was observed through HE staining of pathological sections;the myocardial interstitial and perivascular collagen deposition was observed by sirius red staining;distribution and content of typeⅠand type Ⅲ collagen was analyzed;the expression levels of type Ⅲ collagen gene in myocardial tissue was assayed by fluorescent RT-PCR.Results:The active sites of Scrophulariaceae could lower myocardial hypertrophy index,reduce the cross-sectional area of the myocardial cells of left ventricle,improve myocardial fibrosis and collagen deposition,and significantly bring down the mRNA expression levels of type Ⅲ collagen.Conclusion:The active sites of Scrophulariaceae have significant inhibition effects on both myocardial cells and interstitial collagen deposition,and the action mechanism of the anti-ventricular remodeling may be related to the inhibition of mRNA expression of type Ⅲ collagen.
