七方胃痛胶囊抗胃溃疡作用的实验研究

[目的]探讨中药七方胃痛胶囊抗胃溃疡的机制,及对提高溃疡愈合质量的应用价值。[方法]用乙酸诱发制作大鼠胃溃疡模型后第3d随机分为生理盐水对照组、雷尼替丁胶囊阳性对照组、七方胃痛胶囊大、小剂量组,分别以生理盐水(10ml/kg)、雷尼替丁胶囊溶液(3mg/ml,10ml/kg)、七方胃痛胶囊混悬液(2g/ml或1g/ml,10ml/kg)灌胃,1次/日。灌胃给药14d后处死。用硝酸还原酶法检测血清NO含量,测量大鼠胃溃疡面积,计算溃疡指数,观测溃疡边缘粘膜组织炎症细胞数、新生血管数,并测定溃疡边缘粘膜组织热休克蛋白(HSP)70的表达。[结果]七方胃痛胶囊组的溃疡指数明显低于雷尼替丁组和生理盐水组;七方胃痛胶囊组的再生粘膜的新生血管数明显高于雷尼替丁组和生理盐水组,固有层炎症细胞浸润数明显低于雷尼替丁组和生理盐水组;七方胃痛胶囊组血清NO含量,溃疡边缘粘膜组织的HSP70含量均明显高于雷尼替丁组和生理盐水组。[结论]七方胃痛胶囊抗胃溃疡的作用机制可能是通过提高血清NO及溃疡边缘粘膜组织HSP70含量,降低溃疡指数与炎症细胞浸润数,增加再生粘膜的新生血管生成而发挥作用。七方胃痛胶囊对于提高溃疡愈合质量有肯定的价值。

Experimental Research on Effects of Qifang WeiTong Capsule on Gastric Ulcer

[Objective]To study the mechanism of Qifang WeiTong (QFWT) Capsule on anti-ulcer, and its application for improving the healing of ulcer.[Methods]The ulcer-model rats were induced by ethanoic acid. Three days later, they were randomly divided into the NS group, the Ranitidine group, the low-dosage QFWT group and the high-dosage QFWT group. Each group was treated with physiological saline 10 ml/kg, Ranitidine capsule 3 mg/ml/,10ml/kg, Qifang WeiTong Capsule 1 or 2 g/ml,10ml/kg respectively by gavage for 14 days. Then the rats were killed to determine the NO content in serum by nitrate reductase method. Meanwhile, the ulcerative area was measured to calculate the ulcerative index; the observation was made to see the inflammatory cells in ulcerative mucous membrane, the regenerative blood vessels, and the HSP70 expression.[Results]When compared with the NS group and the Ranitidine group, the ulcerative index and the inflammatory cells in the QFWT groups were significantly lowered; but the regenerative blood vessels and the content of serum NO were obviously increased.[Conclusion]The mechanism of Qifang WeiTong Capsule on anti-ulcer is related to the action of promoting the content of serum NO and the expression of HSP70 as well as increasing the regenerative blood vessels and decreasing the ulcerative index and inflammatory cells. It is effective on improving the healing of ulcer.
