Etest法和NCCLS微量法检测念珠菌对氟康唑的敏感性

摘    要:目的观察氟康唑对念珠菌的敏感性,并研究酵母菌的体外抗真菌药敏试验Etest法和美国国家临床试验标准化委员会(NCCLS)微量法的可比性。方法采用Etest法和NCCLS推荐的药敏试验方案的微量稀释法测定30株念珠菌对氟康唑M IC值。结果采用Etest法和NCCLS微量法测定6株氟康唑耐药株对氟康唑的最小抑菌浓度(M IC)值范围分别均>256μg/m l和>64μg/m l,23株临床分离株对氟康唑的M IC值的范围分别为0.05~2μg/m l和0.125~1μg/m l。Etest法和NCCLS微量法测定30株念珠菌对氟康唑的M IC一致率为83.3%。结论体外抗真菌药敏试验Etest法和NCCLS微量法有较好的一致性、重复性,Etest法可作为一种较好的体外抗真菌药敏试验方法在临床实验室推广使用。

Abstract:Objective To compare the Etest and microdilution methods according to the National Committee for Clinical Laboratory Standards(NCCLS) for determining the minimum inhibitory concentration(MIC) of fluconazole.Methods The MIC of Flucanazale was determined by the Etest and microdilution methods according toNCCLS.The tested strains included 6 FCZ resistant strains,23clinical isolates of Candida albians and 1 standard strains.Results 83.3%agreement of the MICs between the the Etest and microdilution methods according to NCCLS with in ±2 dilution for fluconazole.Of the 6 isolates resistant to fluconazole,the MICs of Flucanazale were >256 μg/ml and>64 μg/ml,of the 23 clinical isolates,the MICs of Flucanazale and itraconazole determined by the Etest were 0.05~2 μg/ml and 0.125~1 μg/ml,were all susceptible to fluconazole.Conclusion The studies indicate that the Etest method is a reproducible and standardized manner.It appears that the Etest method may have clinical utility in laboratories in antifungal susceptibility testing of yeast.
