| 摘 要： | 目的：探讨野生型p53基因对HL－60细胞的作用机制，并检测p21的表达，探讨二者在白血病中的关系。方法：用脂质体介导的方法将野生型p53基因导入p53基因严重缺失的人髓细胞白血病细胞系HL－60中，用形态学，电镜，流式细胞仪，间接免疫荧光等方法检测p53对HL－60细胞的促凋亡作用及对p21表达的调节作用。结果：野生型p53基因能促进HL－60细胞凋亡及上调p21表达。结论：p21可能在p53促HL－60细胞凋亡中起协同作用。 |

| Abstract: | Objective:To study the effect and its mechanism of wild type p53 gene on HL-60 cells and detect the expression of p21 and their relationship with leukemia.Methods:By using lipofectin mediated DNA transfection,wt-p53 was successfully introduced into HL-60 cells.The methods of morphology,electromicroscope,flow cytometry and immunofluorescence were used to detect the proapoptotic effect of wt-p53 on HL-60 cells and its effect of regulating expression of p21.Results:Wild type p53; can induce apotosis of HL-60 cells and upregulate the expression of p21.Conclusion:p21 may cooperate with p53 in the process of p53 inducing apoptosis on HL-60 cells. |
