| 摘 要： | 目的研究结核分枝杆菌gyrA基因突变的规律。方法通过聚合酶链反应-单链构象多态性(PCR-SSCP)技术鉴定106例治疗前后的临床分离株,确定为结核分枝杆菌的菌种,并进一步分析gyrA基因突变的情况。结果以结核分枝杆菌标准株为对照,106株临床分离株的16SrDNASSCP电泳图谱均与结核分枝杆菌标准株相同。59株耐喹诺酮类药临床分离株中30株异常,占50.8%。结论结核分枝杆菌耐药与gyrA基因突变有关,且在药敏实验高、低浓度区均可发生基因突变。

| Abstract: | Objective To study relevance of the mutations of quinolones gyrA genes in M.tuberculosis.Methods 156 clinical isolates were identified of their mycobacterial species,and then analyzed of their gyrA genes with PCR-SSCP.Results 106 clinical isolates had the same SSCP profiles as M.tuberculosis.Of 59 quinolone resistant isolates,30(50.8%)displayed abnormal gyrA SSCP profiles.Conclusion Quinolones resistances in some M.tuberculosis isolates are due to mutations on gyrA genes.Moreover,the mutation of gyrA genes ... |
