摘    要:目的 测定生物检材中安定的含量 ,提高安定的提取率。方法 兔肝经糜蛋白酶水解、固相萃取法提取后 ,用气相色谱 质谱联用仪分析安定的含量。结果 酶解后的肝脏中安定的检出量明显增加 ,每克肝酶用量为 10 0 0U时效果最佳。结论 糜蛋白酶的应用提高了生物检材中安定的提取率

Abstract:Objective To determine the contents of d iazepam in biological samples and to improve its rate of extraction. Methods The samples were hydrolyzed by chymortrypsin of dif ferent quantity,and diazepam was extracted by solid phase columns and its conte nts were analyzed by gas chromatography mass spectrometry(GC MS). Re sults It is shown that the contents of diazepam increased when amount of ch ymotrypsin was increased in the same sample. Conclusions A difficult problem was solved in which diazepam could not be extracted because of its combination with proteins in vitro.This me thod had raised the extraction rate of diazepam in biological samples.
