荧光图像分析法检测树突状细胞在人类肾炎肾组织分布及应用价值

摘    要:通过对荧光双标记染色样本的图像分析,探讨树突状细胞(DC)在肾炎患肾组织中的分布定位及临床意义。方法:选择经肾活检和临床资料确诊的不同类型肾炎患133例。采用免疫荧光双标记染色与低照度荧光图像分析检测方法,观察DC在肾炎患肾组织中分布与改变、结果:肾炎患肾组织中DC分布的面积、数量及密度均明显增多。从分布特点来看,DC以肾小管间质为主,分布于肾小管、肾间质和肾血管,而肾小球基本未见分布,此外,DC在肾小管间质分布程度随肾炎肾小管间质病变程度加重而明显增加,两呈正相关。结论:DC可能参与肾脏疾病的免疫病理机制,并与肾小管间质病变密切相关。

Abstract:Objective:To evaluate the distribution of dendritic cells in tis sue of nephritis human kidney by fluorescence image analysis of double labeling sample.Methods:Biopsy specimen of133patients with nephritis and10normal individ uals were investigated by double immunostaining method and the images were analy zed with Axioplan2microscopy.Results:There were much more double-stained cells in renal tissue of nephritis than that in the normal kidney.In renal tissues of patients with nephritis,dendritic cells were found in renal tubules,interstitiu m and vessels but not glomeruli.The distri-bution area,number and density of de ndritic cells in renal tissues of severe tubulo-interstitial lesion group were greater than those of mild and moderate groups.Conclusions:It is demonstrated t hat mature dendritic cells exist into the kidney with nephritis,which is associa ted with tubulo-interstitial lesions,suggesting antigen-specific immune respon ses may play an important role in the pathogenesis of nephritis.
