结肠腺瘤组织Ezrin蛋白和诱导型一氧化氮合酶表达及其生物学意义的探讨

摘    要:目的探讨Ezrin蛋白、诱导型一氧化氮合酶(iNOS)在结肠腺瘤组织中的表达及其与腺瘤癌变的关系。方法应用免疫组织化学SP法检测60例结肠管状腺瘤、40例绒毛状及绒毛管腺瘤、30例腺瘤癌变中Ezrin蛋白、iNOS和Ki-67核抗原的表达及细胞定位。结果Ezrin蛋白、iNOS在结肠管状腺瘤[58.3%(35/60)、50.0%(30/60)]、绒毛状及绒毛管腺瘤[72.5% (29/40)、70.0%(28/40)]、腺瘤癌变[96.7%(29/30)、93.3%(28/30)]中的表达分别依次上升,差异有统计学意义(Ezrin蛋白:x2= 11.600,P= 0.005;iNOS:X2= 8.451,P= 0.015),Ezrin蛋白与iNOS表达有相关性(r=0.765,P<0.01);Ki-67核抗原增殖指数随Ezrin蛋白、iNOS表达上升而增加。结论Ezrin蛋白和iNOS的过表达可能与结肠腺瘤的增殖、恶性转化有关。

Abstract:Objective To investigate the expression of Ezrin and inducible nitric oxide synthase (iNOS) and their correlation to malignant proliferation of colonic adenoma. Method Expressions of Ezrin,iNOS and nuclear Ki-67 antigen were detected by immunohistochemistry in tubular adenoma (60 cases),villous adenoma (40 cases) and malignant (30 cases ) pleomorphic adenoma. Results Expressions of Ezrin,iNOS in tubular adenoma [58.3% (35/60), 50.0% (30/60)], villous adenoma [72.5% (29/40), 70.00(28/40)]and malignant pleomorphic adenoma [96.7%(29/30),93.3%(28/30)]were gradually increased ( X2 = 11.600,P = 0.005; X2 = 8.451, P = 0.015 ). There was a significant positive correlation between the expression of Ezrin and iNOS (r = 0.765, P < 0.01 ). Nuclear Ki-67 antigen proliferation index was increased with the increasing of Ezrin and iNOS expressions. Conclusion Overexpression of Ezrin and iNOS may promote proliferation and malignant transformation of colonic adenoma.
