小鼠衰老与脑缺血时脑细胞线粒体DNA片段的缺失

目的观察Ｂａｌｂ／ｃ小鼠实验性脑缺血时大脑线粒体ＤＮＡ（ｍｔＤＮＡ）片段的缺失情况。方法用聚合酶链反应（ＰＣＲ）技术进行检测。结果１２只中青年鼠脑细胞未检出ｍｔＤＮＡ片段缺失，检出率为零；８只老年鼠有４只鼠脑细胞检出ｍｔＤＮＡ片段缺失，检出率为５０％。当脑缺血后，此种片段缺失率明显增高，１７只中青年鼠检出率为７０．６％，１１只老年鼠检出率为１００％。结论脑细胞ｍｔＤＮＡ片段缺失检出率随鼠龄增长而增加，也随脑缺血而增加。

Mitochondrial DNA deletions in aging and ischemic Balb/c mouse brain

Objective To detect the deletion of mitochondrial DNA (mtDNA) from ischemic cerebral cells of Balb/c mouse. Methods By using polymerase chain reaction technique and electrophoresis to examine the fragment deletion of mtDNA. Results The deletion rate of mtDNA in brain of 12 young mice was nought (0/12), but 50% (4/8) in old mice. After brain ischemia, the deletion rate of mtDNA increased to 70 6% (12/17) in young mice, and 100% (11/11) in old mice. Conclusions The deletion rate of mtDNA in brain increased with ageing and cerebral ischemia.
