补中益气汤含药血清干扰A549/DDP生长作用的实验研究

目的从细胞水平探讨补中益气汤含药血清改善A549/DDP耐药作用并选择最佳含药血清的浓度。方法采用随机对照法将24只SD大鼠分为高、中、低剂量组和空白对照组,制备高、中、低剂量补中益气汤的含药血清,分别以5%和10%的浓度作用于人肺腺癌耐药细胞株A549/DDP 48 h,观察补中益气汤含药血清对A549/DDP生长的影响并通过MTT法检测各组细胞的抑制率,以此筛选出最佳含药血清浓度,计算出最佳浓度含药血清对A59/DDP的耐药逆转倍数。结果补中益气汤含药血清对A549/DDP细胞有明显抑制作用,且呈浓度依赖关系。5%高剂量组对A549/DDP的抑制率高于5%中剂量组和5%低剂量组,并与5%空白血清组比较,差异有统计学意义（P〈0.05）。10%各剂量组对A549/DDP的抑制率与10%空白血清组比较,差异均有统计学意义（P〈0.05）。经筛选,10%中剂量含药血清作用48 h为最佳干扰浓度,在此浓度下,A549/DDP对顺铂的IC50明显下降（P〈0.05）,对A549/DDP细胞的逆转倍数为2.46。结论 10%中剂量含药血清干扰48 h可作为后期药物实验的含药血清加药浓度及观察时间筛选的实验依据,补中益气汤含药血清可逆转A549/DDP的耐顺铂作用。

Research on efficiency of Buzhong Yiqi Decoction medicated serum on A549/DDP cell

Objective To analyze the reversal effect of Buzhong Yiqi Decoction medicated serum on A549/DDP mul tidrug resistance,and choose the best drug level.Methods The randomized controlled method was used to divide the 24 SD rats into the high,middle,low dose of Buzhong Yiqi Decoction group,and the blank group,Drug-containing serum was prepared,then the effects of 5% and 10% serum of each group for 48 h on A549/DDP cells proliferaton were observed by MTT assay,in order to select the best containing medicine serum concentration,then the reversal fold of optimal concentration medicated serum on A59/DDP was calculated.Results Buzhong Yiqi Decoction inhibited the growth of 549/DDP obviously,and it was observed to exert a concentration-dependent inhibition on the prolifera tion of A549/DDP cell line.The A549/DDP suppression ratio in 5% high dose group was higher than 5% middle dose group and 5% low dose group,and compared with the 5% blank group,the difference was statistically significant（P ＆lt; 0.05）.The A549/DDP suppression ratio in 10% high,middle and low dose groups were statistical differences with 10% blank group（P ＆lt; 0.05）.The best concentration of Buzhong Yiqi Decoction medicated serum was 10% middle dose of serum for 48 h,under this concentration,the IC50 of Cisplatin decreased（P ＆lt; 0.05）,the reverse rate was 2.46.Conclusion 10% middle dose of Buzhong Yiqi Decoction medicated serum working 48 h can serve as experiment based on the late drug tests,in the aspects of medicated serum drug concentration and observe time,and it can moderately re verse the multidrug resistance of A549/DDP cells for Cisplatin.
