竹节参总皂苷的制备工艺及含量测定

目的：优选竹节参总皂苷的提取纯化工艺，并测定其含量。方法：运用正交试验设计，以药材中总皂苷的转移率作为考察指标，筛选最佳提取工艺；用正丁醇萃取合并醇溶丙酮沉淀法纯化得到精制皂苷；分别采用分光光度法和HPLC测定其中总皂苷和三萜苷元齐墩果酸的含量。结果：提取工艺以药材粗粉10倍量的60%乙醇，浸泡2 h后加热回流提取3次，每次3 h最佳；纯化工艺中以正丁醇萃取3次，以85%乙醇充分溶解后，边搅拌边缓慢加入4～5倍量丙酮沉淀效果最佳。经纯化后总皂苷可达8348%，其中三萜苷元齐墩果酸可达3830%。结论：本工艺条件对竹节参总皂苷的提取收率高且稳定，测定方法简便可行，重现性好。

Studies on preparative technology and quantitative determination for extracts of total saponin in roof of Panax japonicus
