摘    要:目的观察cripto-1基因小于扰RNA(siRNA)对乳腺癌细胞侵袭力的影响。方法将人乳腺癌MDA—MIM68细胞分为4组,A组不处理,B组转染空载脂质体,C组转染错配siRNA,D组转染分别转染含3.125、6.25、12.5nmol/Lcfipto-1基因siRNA的脂质体。分别于处理24.48、72h收集各组细胞。采用实时定量RT—PCR法和Western blot法检测MDA—MB468的cripto-1 mRNA和蛋白,采用软琼脂集落培养试验和Boyden小室检测MDA—MB468的锚着不依赖性增殖和侵袭能力。结果与A、B、C组比较,D组MDA—MB468的cripto-1 mRNA、cripto-1蛋白、集落形成数量、穿膜细胞数均呈浓度依赖性减少(P均〈0.005)。结论cripto-1基因siRNA可抑制乳腺癌细胞侵袭。
