多肽P161联合顺铂对乳腺癌MA-782细胞株增殖和凋亡的影响

摘    要:目的:探讨特异性多肽P161与顺铂(CDDP)联合应用时乳腺癌细胞株MA-782增殖及凋亡的影响。方法:应用四甲基偶氮唑比色法(MTT)测定P161、CDDP单用及联合应用对MA-782细胞增殖的抑制,光学显微镜观察细胞形态学改变,荧光显微镜观察经Hoechst33342染色后细胞凋亡形态改变,流式细胞术检测细胞凋亡率。结果:MTT显示,与单独用药组相比,联合用药组细胞增殖活力显著降低,呈剂量依赖性,且与CDDP组比较,差异有统计学意义(P〈0.05);倒置显微镜观察,与单独给药组相比,联合用药组细胞形态改变愈加明显;Hoechst33342染色显示,与单独用药组相比,联合用药组凋亡形态特征更为显著,染色质凝集现象增多。流式细胞术检测显示,与CDDP组相比,联合给药组细胞早期凋亡率显著升高,其凋亡率由(30.03±0.93)%增至(61.67±0.81)%(P〈0.01)。结论:多肽P161联合CDDP可增加乳腺癌细胞株对CDDP的敏感性,此作用可能与促进细胞早期凋亡有关。

Abstract:Objective:To explore the effect of polypeptide P161 combined with cisplatin on the proliferation and apoplosis of breast cancer cell line MA-782.Method:PI61 and CDDP were added into the cultured carcinoma cell line MA-782 in different ways. After 24 hours,MTT was used to determine the combinative effect,and the morphological changes of MA-782 cells were observed under an inverted microscope.The apoplosis of cells after adding different concenlralions of drugs was observed by Hoechst33342 fluorescence staining and flow cytometry.Result;The results of MTT showed the combination group could significantly decrease cell proliferation aclivily in a dose-dependent manner compared with the single group,and compared with CDDP group,the difference was statistically significant(P <0.05).Hoechst33342 staining showed the combination group had more obvious chromatin condensation phenomena compared with the single group.The combination group showed much higher early apoplosis rate than the single group delected by flow cytomelry(P<0.05),and the apoplosis rate was increased from(30.03 ±0.93)%to(61.67 ±0.81)%(P<0.01).Conclusion;CDDP combined wild PI61 in chemotherapy can synergistically inhibit the proliferation of MA-782 cells,and significantly increase the chemo-sensitivity of MA-782.The function may be associated will) the promotion of early apoptosis.
