KAI1和nm23与人黑色素瘤细胞转移潜能的相关性

摘    要:目的:探讨肿瘤转移抑制基因KAI1和nm23与人黑色素瘤细胞转移潜能的关系.方法:应用Northern blot杂交检测不同转移潜能人黑色素瘤细胞中KAI1和nm23基因的表达.结果:4种不同转移潜能的人黑色素瘤细胞中(WM35,WM451,WM983a,WM1341b),KAI1 mRNA和nm23 mRNA均有表达且表达量基本一致,KAI1在4种细胞中的积分光密度值分别为0.1798,0.1800,0.1501,0.1582;nm23分别为1.0428,1.0607,1.0141,1.0754.结论:KAI1基因与nm23基因的表达降低与人黑色素瘤的浸润转移无关.

Abstract:Aim:To investigate the correlation of tumor metastasis suppressor gene KAI1, nm23 with metastatic potential of human melanoma cell lines Methods:Expressions of KAI1 gene and nm23 gene in human melanoma cell lines with different metastasis potential were studied by Northern blot hybridization Results:KAI1 mRNA and nm23 mRNA all expressed in four human melanoma cell lines (WM35, WM451, WM983a, WM1341b) and no difference in expression levels were observed in these cancer cell lines(integral light density values in KAI1 gene are 0 179 8, 0 180 0, 0 150 1, 0 158 2 respectively and in nm23 gene are 1 042 8, 1 060 7, 1 014 1, 1 075 4 respectively) Conclusion:Reduced KAI1 and nm23 expressions may be no association with enhanced potential in human melanoma cells
