RP-HPLC测定六味木香散中胡椒碱含量

 目的:建立六味木香散中胡椒碱含重测定方法。方法:反相液相色谱法。采用Shim-Pack CLC-ODS(15 cm X6 mm)分析柱;ODS保护柱;流动相:甲醇-水(67:33);流速:1.0 ml · min^-1;色语柱温度35℃;检测波长:343 nm;进样体积10μl;外标法计算含量。结果:理论板数按胡椒碱峰计算为3 600;分离度为2.5;胡椒碱浓度在3～48μg·ml^-1范围与色谱峰面积呈线性关系，r = 0.999 9;回收率及RSD分别是98.6%和2.1%。结论:方法简便准确，可用于六味木香散中胡椒碱含量测定。

Determination of piperine in Liuwei Muxiang San by RP HPLC

OBJECTIVE: To establish a quantitative method of piperine in Liuwei Muxing San(Chinese petent medicine).METHODS: Piperine in Liuwei Muxiang San was determinated by RP-HPLC. Chromatographic conditions included column ODS-C₁₈, column temperature, 35℃;detector UV, 343 nm; mobile phase, methanol-water(67:33); flow rate, 1 ml·min^-1;injecting volume, 10μl; external standard. RESULTS: The number of theoretical plates calculated bY piperine peak was 3 600. The resolution was 2.5. The standard curve was linear in the concentration range of 3～48μg· ml^-1，and correlation coefficient was 0.999 9. The average recovery and the relative standard deviation were 98.6 % and 2.1%，respectivily. CONCLUSION; The method is simple, accurate, and it is satisfactory for determination of piperine in Liuwei Muxiang San.
