肺气虚证大鼠血液流变学指标与内皮素、血栓素水平的相关性

摘    要:背景肺气虚证的实质近年来得到较深入的研究,但其客观定量化标准的确立仍具有很大困难,很难为之确定一个或几个相关性很强的客观指标.目的研究肺气虚证大鼠的血液流变学变化与内皮素、血栓素B2变化的相关性,为肺气虚证客观定量化标准提供实验依据.设计随机对照实验研究.地点和材料60只大鼠随机分为模型组与对照组,每组30只;实验在安徽医科大学核医学室和安徽中医学院附属医院实验中心完成.干预模型组复制肺气虚证模型,对照组置于正常无烟环境.主要观察指标全血黏度(高切、低切)、血浆黏度、红细胞比容、红细胞聚集指数及纤维蛋白原、ET和TXB2.结果模型组与对照组比较,全血黏度(高切、低切)、血浆黏度、红细胞比容、红细胞聚集指数及纤维蛋白原均升高,有显著性差异(P<0.01或P<0.05);模型组内皮素和血栓素B2明显升高,与对照组相比,差异有显著性(P<0.01).模型组全血黏度与内皮素、血栓素B2呈正相关(r=0.957,P<0.05;r=0.992,P<0.01).结论肺气虚证大鼠存在"血瘀"现象,且血浆中的内皮素、血栓素B2含量随肺气虚证逐渐加重而呈上升趋势.

Abstract:BACKGROUND: The essence of lung qi deficiency syndrome has been Penetratingly studied in recent years, but it is difficult to establish an objective and quantified standard as well as to set several closely correlated objective indices.OBJECTIVE: To study the correlation between hemorheology indices and the levels of thromboxane B2(TXB2) and endothelin(ET) in rats with lung qi deficiency syndrome to provide a laboratorial gist for the establishment of the objective quantified standard in lung qi deficiency syndrome.DESIGN: A randomized controlled trial.SETTING and PARTICIPANTS: Sixty rats were randomly allocated into control group and model group with 30 each. Study was conducted in the Department of Nuclear Medicine of Anhui Medical University and the Experimental Center, Affiliated Hospital of the Anhui Traditional Chinese Medicine College .INTERVENTIONS: Lung qi deficiency syndrome model was established in the rats of model group, and rats of control group were left in normal smokeless environment.MAIN OUTCOME MEASURES: The whole blood viscidity (high-shearing and low-shearing), plasma viscidity, hematocrit, erythrocyte aggregation index, fibrinogen, ET and TXB2.RESULTS: The whole blood viscidity(high-shearing, low-shearing), plasma viscidity, hematocrit, erythrocyte aggregation index and fibrinogen all increased in model group compared with that of control group( P<0.05-0.01 ), and the levels of ET and TXB2 increased significantly compared with that of control group( P<0.01) . The whole blood viscidity was positively correlated with ET and TXB2 respectively(r= 0.957, P<0.05; r = 0. 992,P < 0.01).CONCLUSION: Lung qi deficiency syndrome has the phenomenon of blood stasis and the levels of ET and TXB2 in plasma tend to elevate with the gradual aggravation of lung qi deficiency syndrome.
