摘    要:目的 :观察肝缺血再灌注对细胞周期的影响 ,以探讨移植肝功能不良的可能原因。方法 :18只Wistar大鼠随机分成假手术组 (sham) ,缺血 1h再灌注 1h组 (IR1) ,缺血 1h再灌注 4h组 (IR2 )三组。利用肝原位部分缺血再灌注模型 ,用流式细胞检测技术定量测定各期细胞数 ,观察随灌注时间的延长细胞周期的变化。结果 :与对照组相比 ,IR1组肝组织的细胞周期S期比例降低 ,G2 /M期比例升高 ;IR4组G0 /G1期细胞比例增加 ,G2 /M期细胞比例明显减少。结论 :肝缺血再灌注能引起细胞周期的变化 ,使细胞的增殖和分裂处于抑制状态 ,从而影响细胞的再生和组织的修复

Abstract:Objective:To observe the effects of hepatic ischemia reperfusion on cell cycle, and to investigate the probable causes of the poor liver function after transplantation. Methods: Eighteen Wistar rats were randomly divided into three groups: Sham operation group (sham group); Ischemia one hour reperfusion one hour group (IR1);Ischemia one hour reperfusion four hours group (IR2). The model of partial liver ischemia/reperfusion was used.The number of cells in each phase was measured by flow cytometry to observe changes in cell cycle as time of reperfusion extending.Results:Compared with sham group, S-phase ratio decreased and G 2/M-phase ratio increased in IR1 group; G 0/G 1-phase ratio increased and G 2 /M-phase ratio decreased significantly in IR2 group.Conclusion:Hepatic ischemia/reperfusion changed cell cycle, cell proliferation and split in rat liver were in restrained state that would influence cell regeneration and repair of tissue.
