摘    要:目的 了解血管内皮生长因子(VEGF)在一过性全脑缺血再灌注鼠脑表达的动态变化。方法 采用Pulisnelli改良法四血管堵塞全脑缺血再灌注模型,用免疫组织化学方法观察了全缺血15min再灌注2-14d时,VEGF表达的动肪变化。结果 全脑缺血15min再灌注6h VEGF即可在大脑皮层,纹状体及丘脑等区域的血管内皮细胞表达,1d达高峰,一直持续到缺血后再灌注3d,结论 脑缺血后再灌注可上调VEGF在大脑皮质,纹状体及丘脑等区域内皮细胞的表达,VEGF可能通过促进血管内皮细胞生长对缺血性神经元损伤起一定的作用。

Abstract:Objective To investigate the time course of changes of vascular endothelial growth factor (VEGF) expression in rat brains following transient global ischemia.Methods The four vessel occlusion ischemic model of Sprague Dawley rats were made for 15 minutes and recirculated for 2h to 14 days. The expression of VEGF was examined by means of immunohistochemistry method.Results Expressions were rapidly induced (with 6 h) in the cerebral cortex, striatum and thalamus by transient global ischemia (15 min), peaked at 1 day, and lasted at least 3 days.Conclusions These findings suggest that induction of VEGF is up regulated by ischemia. VEGF may play an important role in post ischemia cerebral injury by the maintenance and induction of growth of vascular endothelial cells.
