| 摘 要： | 目的探讨菟丝子含药血清对体外培养大鼠成骨细胞代谢调控的影响。方法采用血清药理学方法制备菟丝子含药血清;采用改良的组织块法体外培养大鼠成骨细胞;分别用5%、10%、15%、20%菟丝子含药血清诱导培养成骨细胞24h、48h、72h后,MTT法检测大鼠成骨细胞的增殖情况;用对硝基苯二钠基质动力学法(PNPP)测定细胞内碱性磷酸酶(ALP)的活性;培养48h后用RT-PCR方法检测Ⅰ型胶原(COL-Ⅰ)mRNA的表达。结果 5%和10%菟丝子含药血清在48h和72h能显著促进成骨细胞增殖并提高ALP活性;在48h时10%菟丝子含药血清可促进COL-ⅠmRNA的表达。结论 10%菟丝子含药血清能够明显促进大鼠成骨细胞的增殖与分化,表明其具有促进成骨作用。
