湿阻中焦证模型胃肠水通道蛋白2的表达分布谱及平胃散的干预作用

摘    要:目的探讨平胃散对湿阻中焦证模型的干预作用机制。方法 SD大鼠50只,随机分空白组、空白给药组、模型组、平胃散组和自然恢复组,每组10只。模拟外湿过盛、饮食不节、情志不遂三大致湿病因,建立湿阻中焦证模型。空白给药组和平胃散组给予平胃散汤剂灌胃10ml/kg,空白组和自然恢复组给予0.9%生理盐水灌胃10ml/kg,3天后取材。用免疫组化技术测定胃肠组织AQP2的分布,用酶联免疫法测定胃肠组织AQP2的含量。结果模型组大鼠AQP2在胃体中间黏膜、大肠中段黏膜下组织表达减少,在小肠中段组织、大肠黏膜、大肠末端黏膜下组织表达增加。平胃散增强湿阻中焦证胃体中间黏膜、大肠中段的黏膜AQP2的表达,抑制湿阻中焦证大肠中段黏膜下组织AQP2的表达。结论 AQP2在湿阻中焦证胃肠特征性表达分布谱可能是湿阻中焦证的分子基础之一;平胃散对其表达的干预可能是该方治疗湿阻中焦证的分子机理之一。
