子宫内膜异位症患者在位内膜分泌RANTES及对单核细胞的趋化活性

摘    要:目的了解子宫内膜异位症(Em)患者的在位内膜在分泌RANTES及对单核细胞的趋化活性是否有别于非Em患者子宫内膜。方法白细胞介素(IL)-1β刺激培养Em与非Em患者子宫内膜基质细胞,分别在刺激培养后12、24、36、48、60、72、84和96 h取上清酶联免疫吸附试验(ELISA)检测RANTES水平;Boyden小室趋化实验分析刺激培养上清液对单核细胞的趋化活性及RANTES趋化单核细胞的能力。结果Em在位内膜从刺激培养60 h始,RANTES分泌量及趋化单核细胞的能力显著高于非Em在位内膜;Em在位内膜基质细胞刺激培养60 h上清所趋化的单核细胞中有55%是由RAN-TES趋化而来。结论在相似的培养条件下,Em患者的在位内膜基质细胞RANTES的表达模式和趋化单核细胞的活性不同于非Em患者子宫内膜。

Abstract:Objectives: To evaluate if the expression and chemotactic activity of RANTES are different in IL-1β treated autologous eutopic endometrial stromal cells from endometriosis patients compared to non-endometriosis eutopic endometrium.Methods: Eutopic endometrial stromal cells from patients with or without endometriosis were collected and cultured.IL-1β was added to induce the secretion of RANTES.Concentrations of RANTES in the culture medium at 12 h,24 h,36 h,48 h,60 h,72 h,84 h and 96 h were measured with a sandwich ELISA.U937 monocyte chemotactic activity was assayed in Boyden Chambers.Results: The RANTES secreted by the stromal cells of autologous eutopic endometrium from endometriosis patients was increased significantly than that from non-endometriosis women after 60 h of culture.The RANTES accounts for the majority(55%) of the monocyte chemotactic activity in autologous eutopic endometrial stromal cells conditioned media. Conclusions: The expression of RANTES and its monocyte chemotactic activity of autologous eutopic endometrium in the endometriosis patients were different significantly from those in non-endometriosis women.
