肉蔻五味丸对大鼠心肌梗死后左室重构的影响

目的探讨肉蔻五味丸对心肌梗死后左室重构大鼠心肌基质金属蛋白酶（MMPs）的影响。方法 SD大鼠36只,随机分为正常对照组、心肌梗死模型组、肉蔻五味丸治疗组。模型组和治疗组大鼠腹腔注射异丙基肾上腺素（IS0）,治疗组给予肉蔻五味丸1g/kg·d灌胃,至各相应时段。称重并计算左室重量指数,免疫组化SABC法测各组不同时段心肌间质MMP-2、MMP-9表达,光镜下观察心肌病理改变。结果模型组大鼠左室重量指数高于正常对照组,心肌间质MMP-2、MMP-9表达在各时段明显高于对照组。肉蔻五味丸组左室重量指数、MMPs活性均显著低于模型组（P〈0.05或P〈0.01）。结论大鼠心肌梗死后心肌间质内的MMP-2、MMP-9表达升高,是心室重构的重要原因之一。蒙药肉蔻五味丸对MMPs活性的抑制可能是其干预治疗心肌梗死后左室重构的机制之一。

Effects of RouKouWuWei Pill on Ventricular Remodeling following Myocardial Infarction in the Rats

Objective To study the effects of RouKouWuWei Pill on matrix metalloproteinases（MMPs） of left ventricular remodeling following myocardial infarction in the rats.Methods Thirty-six SD rats were randomly divided into normal control group、myocardial infarction group and Mongolian drug group.The rats of myocardial infarction group and drug-treated groups were given isoproterenol（ISO） by ip,subsequently drug-treated groups given Mongolian drug 1 g/kg·d by ig until this experiment terminal.Left ventricular weight indexes were calculated and the expression of MMPs were measured by SABC.The pathologic change of myocardium was observed with microscope.Results Left ventricular weight index and the expression of MMP-2,MMP-9 in the myocardial infarction group increased significantly compared with normal control.The weight index and the activities of MMPs decreased in Mongolian drug group compared with myocardial infarction group.（P0.05或P0.01）.Conclusion The expression of MMPs in the interstitium increased in some trends following myocardial infarction,and this change may be the major cause of ventricular remodeling.The effects of Mongolian Drug RouKouWuWei Pill on the activities of MMPs may be one of mechanisms on the ventricular remodeling following myocardial infarction.
