针刺对金黄地鼠下丘脑腹内侧核和背内侧核一氧化氮合酶表达的影响

摘    要:目的观察针刺对金黄地鼠下丘脑腹内侧核(VMH)和下丘脑背内侧核(DMH)中神经元型一氧化氮合酶(nNOS)表达的影响。方法将20只健康雄性叙利亚金黄地鼠随机分为4组,每组5只。金黄地鼠经驯化后进入自由运行状态,在恒定黑暗第3天于近似昼夜时间(CT)9:00分别给予捆绑对照、8-羟基-丙胺-四氢柰(8-OH—DPAT)、光脉冲和电针刺激。2小时后用免疫组化方法检测各组动物VMH和DMH中nNOS阳性细胞并对此进行计数。采用ANOVA方法进行统计学分析。结果与对照组比较,针刺组VMH和DMH中nNOS阳性神经元数量均明显增加,光照组和8—0H—DPAT则无明显变化。结论针刺在主观白天CT9能影响VMH和DMH内nNOS的表达。

Effect of acupunctrure on the expression of nitricoxide synthase in ventromedial hypothalamic nucleus and dorsomedial hypothalamic nucleus of golden hamsters

Abstract:Objective To observe the effect of electroacupuncture on the expression of neuronal nitricoxide synthase (nNOS) in ventromedial hypothalamic nucleus (VMH) and dorsomedial hypothalamic nucleus (DMH) of golden hamsters. Methods A total of 20 male Syria golden hamsters were divided randomly into 4 groups, each containing 5 samples. They moved freely following domestication. At circadian time (CT) 9:00 on the third day of constant darkness, animals were treated by seizing, 8-hydroxy-2- (di-n-propylamino) tetralin (8-OH-DPAT),photic stimuli and elecroacupunctrure respectively. Two hours later, immunohistochemistry was utilized to observe the expression of nNOS in VMH and DMH. And nNOS immunoreactive neurons was counted. We used ANOVA to test intergroup differences.Results Comparing with seizing control group,the expression of nNOS in electroacupunctrure group increased obviously in VMH and DMH. No obvious change was seen in photic and 8-OH-DPAT groups. Conclusion Acupunctrure can affect expression of nNOS in VMH and DMH at subjective day.
