汉防己乙素对大鼠肝脏缺血再灌注损伤脂质过氧化动态变化的影响

摘    要:目的 观察汉防己乙素对大鼠肝脏缺血再灌注损伤时脂质过氧化动态变化的影响。方法 制作大鼠肝脏缺血再灌注损伤模型,随机分为假手术组、缺血再灌注组及汉防己乙素低、高剂量预处理组,每组10只。连续观察各组缺血前、缺血30 min、再灌注30 min、1 h及2 h血清中丙氨酸氨基转移酶(ALT)、超氧化物歧化酶(SOD)、丙二醛(MDA)活性的动态变化,并观察汉防己乙素预处理组对各项指标的影响。结果 缺血再灌注组各时点血清ALT、MDA呈逐渐升高的变化,SOD呈逐渐降低的改变;肝组织MDA、SOD变化各组均有差异;汉防己乙素组与缺血再灌注组比较差异均有显著或非常显著意义(P<0.05或P<0.01)。结论 肝脏缺血再灌注损伤时脂质过氧化呈进行性加重的变化;汉防己乙素能抑制氧自由基的生成,增强对其的清除,具有较好的保护作用。

Abstract:Objective This study was designed to investigate the effect of demethyl-tetrandrine(d-Tet) on dynamic changes of lipid peroxidation in hepatic ischemia-reperfusion injury (HIRI). Methods 40 rats were randomly divided into four groups (n = 10) including sham-operation groups, ischemia reperfusion(I/R)groups, 7.5mg/kgand 15mg/kg d-Tet Preconditioning(L-dTPC and H-dTPC)groups by structuring the HIRI model. The alanine aminotransferase (ALT) activity,malondialdeyde (MDA) content and superoxide dismutase (SOD) activity in serum were observed for 30 minutes during pre-ischemia and ischemia,another session for reperfusion (30 min, 1h and 2h). Meanwhile, MDA and SOD in hepatic tissue were determined in reperfusion (2h) after ischemia. Results With continuum of ischemia-reperfusion, ALT、MDA elevated and SOD reduced remarkably step by step in I/R groups. There existed differential changes of MDA content and SOD activity in hepatic tissue during reperfusion (2h) after ischemia. All above parameters of d-Tet groups, compared with I/R groups, showed significant differences during the session of reperfusion . Conclusion D-Tet has notable anti-lipid peroxidation effect on HIRI by inhibiting oxygen free radicals generation and strengthening scavenging oxygen free radicals.
