流式细胞术测定血小板活化在先兆子痫孕妇中的临床应用

目的运用流式细胞术测定血小板的活化状态，探讨先兆子痫孕妇体内静息态与活化态血小板CD41a、CD62p、CD63等血小板膜糖蛋白表达量的变化及其临床意义。方法用免洗的全血免疫直标法，将血小板CD41a、CD62p、CD63荧光单抗分别标记25例正常人与20例正常孕妇及22例先兆子痫孕妇静息态及ADP活化后血小板，在流式细胞仪上测定其表达量。结果正常孕妇组与正常对照组比较，静息态间及活化态间血小板膜糖蛋白CD62p、CD63、CD41a表达量都没有显著性差异；而先兆子痫孕妇组静息态与活化态血小板膜糖蛋白CD62p、CD63表达量均较正常孕妇组显著增高，CD41a表达无显著性差异。结论先兆子痫产妇体内血小板膜糖蛋白CD62p、CD63表达处于较高水平，提示血小板处于活化状态。

The Detection of Platelete Activity by Flow Cytometry Method in the Gravida with Pre-eclampsia

Objective The activation of platelete was detected by using the flow cytometry method te-chonolygy. To explore the clinic significance of the variation of the platelete glycoprotein CD 41a,CD 62p,CD 63 expression in static and activated in the gravida with pre-eclampsia. Methods We used no-washed immunity labelling technique in blood of subjects which included 16 gravida with pre-eclampsia and 15 normal pregnancies and 20 health persons. The fluorescence labblled monoantibodies of platelete glycoprotein included CD 41 a,CD 62p,CD 63 were reacted with static and activated platelete. The expression of monoantinbody were dectected by flow cytometry. Results The average static and activated platelete glycoprotein CD 41 a,CD 62p, CD 63 expression levels in the gravida with pre-eclampsia were apparently higher than those of the normal pregnancy (P <0. 01 ) ; while the comparison between the latter and the control group did not reveal significant difference (P >0. 05). Conclusion High levels of platelete glycoprotein CD 62p and CD 63 expression are present in gravida with pre-eclampsia. It is infer the activation of platelet were present in the gravida with pre-eclamsia.
