CO_2对卵巢癌细胞株SKOV3抗凋亡因子XIAP和PED/PEA-15表达的影响

摘    要:目的:探讨CO_2气腹对卵巢癌细胞株SKOV3抗凋亡因子XIAP和PED/ PEA-15表达的影响。方法:建立体外模拟CO_2气腹环境,将人卵巢癌细胞株SKOV3分别暴露于不同压力(8~12mmHg)的CO_2环境中,持续1、2、3h后,脱离CO_2环境,放入常规CO~2培养箱中培养,分别于培养24、48、72h后用实时荧光定量PCR法及免疫细胞化学法检测各组SKOV3细胞XIAP、PED/PEA-15 mRNA和蛋白的表达。结果:SKOV3细胞置于不同的CO_2气腹环境后,与对照组比较,SKOV3细胞XIAP、PED/PEA-15的表达明显增加(P<0.05),并且与CO_2压力和作用时间有关。结论:CO_2气腹环境可促进SKOV3细胞中抗凋亡因子XIAP、PED/PEA-15表达增加,从而抑制细胞凋亡。

Abstract:Objective:To explore the effects of carbon dioxide on the expressions of antiapoptosis factors XIAP and PED/PEA-15 in human ovarian cancer cell line SKOV3.Meth- ods:Human ovarian cancer cell line SKOV3 was put in analogous CO_2 pneumoperitoneum envi- ronment.Realtime FQ-PCR and immunocytochemical staining were used to examine the expres- sions of XIAP and PED/PEA-15 of these cells after exposure to different pressures and persist- ence of CO_2.Results:CO_2 could significantly increase the expressions of XIAP and PED/PEA- 15(P<0.05).The expressions of XIAP and PED PEA-15 were related with the pressure and persistence of CO_2.Conclusion:CO_2 can suppress apoptosis capacity of SKOV3 cell by upregu- lating the expressions of antiapoptosis factors XIAP and PED/PEA-15.
