缺氧对口腔癌细胞增殖和凋亡的作用

摘    要:目的检测口腔癌细胞中缺氧情况及缺氧/复氧对口腔癌细胞增殖、凋亡的影响。方法设立对照组和不同时间缺氧/复氧6个实验组,采用细胞培养、缺氧探针、MTT法及流式细胞术等方法检测SCC15口腔癌细胞中缺氧情况及缺氧/复氧对口腔癌细胞增殖、凋亡的影响。结果对照组和实验组SCC15细胞均呈现缺氧状态,随着缺氧时间的延长,活细胞数减少,细胞形态呈现细胞凋亡改变。缺氧12h组、24h组、缺氧12h/复氧2h组、缺氧24h/复氧2h组细胞增殖被显著抑制,随缺氧时间延长,SCC15细胞生长抑制的比例逐渐加大(P0.01)。缺氧对不同实验组SCC15细胞均能促进细胞凋亡。随缺氧时间延长,SCC15细胞发生凋亡的比例逐渐增高。除缺氧4h组外,其它各组与对照组相比,差异具有统计学意义((P0.01)。结论缺氧与口腔癌细胞增殖和凋亡密切相关,缺氧程度影响着口腔癌细胞的生长。

Abstract:Objective To investigate the hypoxia and its impact on the proliferation and apoptosis in oral cancer cells.Methods The hypoxia,growth and apoptosis in SCC15 oral cancer cell were observed by Hypoxyprobe-I MabI,MTT assay,flow cytometry in the control and six groups with different time of hypoxia/reoxygenation.Results Hypoxia was found in the control and hypoxia/reoxygenation groups.The percentage of viable cells was steadily reduced in hypoxia-treated groups.Hypoxic growth induced morphological alterations typical for apoptotic cell death.Hypoxia inhibited cell growth and induced cell apoptosis in SCC15 cells after 12h or 24h hypoxia/reoxygenation(P0.01).Conclusion Hypoxia inhibits the cell proliferation and promotes apoptosis in oral cancer cells.The extent of hypoxia has effects on oral cancer cell growth and apoptosis.
