缺氧对口腔癌细胞增殖和凋亡的作用

目的检测口腔癌细胞中缺氧情况及缺氧/复氧对口腔癌细胞增殖、凋亡的影响。方法设立对照组和不同时间缺氧/复氧6个实验组,采用细胞培养、缺氧探针、MTT法及流式细胞术等方法检测SCC15口腔癌细胞中缺氧情况及缺氧/复氧对口腔癌细胞增殖、凋亡的影响。结果对照组和实验组SCC15细胞均呈现缺氧状态,随着缺氧时间的延长,活细胞数减少,细胞形态呈现细胞凋亡改变。缺氧12h组、24h组、缺氧12h/复氧2h组、缺氧24h/复氧2h组细胞增殖被显著抑制,随缺氧时间延长,SCC15细胞生长抑制的比例逐渐加大(P0.01)。缺氧对不同实验组SCC15细胞均能促进细胞凋亡。随缺氧时间延长,SCC15细胞发生凋亡的比例逐渐增高。除缺氧4h组外,其它各组与对照组相比,差异具有统计学意义((P0.01)。结论缺氧与口腔癌细胞增殖和凋亡密切相关,缺氧程度影响着口腔癌细胞的生长。

Influence of hypoxia on proliferation and apoptosis in oral cancer cells

Objective To investigate the hypoxia and its impact on the proliferation and apoptosis in oral cancer cells.Methods The hypoxia,growth and apoptosis in SCC15 oral cancer cell were observed by Hypoxyprobe-I MabI,MTT assay,flow cytometry in the control and six groups with different time of hypoxia/reoxygenation.Results Hypoxia was found in the control and hypoxia/reoxygenation groups.The percentage of viable cells was steadily reduced in hypoxia-treated groups.Hypoxic growth induced morphological alterations typical for apoptotic cell death.Hypoxia inhibited cell growth and induced cell apoptosis in SCC15 cells after 12h or 24h hypoxia/reoxygenation（P0.01）.Conclusion Hypoxia inhibits the cell proliferation and promotes apoptosis in oral cancer cells.The extent of hypoxia has effects on oral cancer cell growth and apoptosis.
