西洋参石斛药膳氨基酸类成分HPLC指纹图谱研究

摘    要:目的:采用柱前衍生-反相高效液相色谱法(RP~HPLC),建立西洋参石斛药膳中氨基酸类成分的指纹图谱。方法:采用2,4-二硝基氟苯柱前衍生化法,色谱柱为Kromasil C18(250ram×4.6ram,5um),流动相A为乙腈,B为N,N-二甲基甲酰胺-0.025tool·L^-1醋酸钠(1:100,体积分数36%乙酸调至pH6.0),梯度洗脱,流速为1.0mL·min^-1,柱温为40℃,检测波长为360nm,进样量为10μL,测定10批西洋参石斛药膳的指纹图谱。结果:标定了23个共有峰,鉴定出17种氨基酸成分,含所有人体必需氨基酸;10批药膳样品指纹图谱的相似度大于0.92,初步建立了西洋参石斛药膳氨基酸类成分的HPLC指纹图谱。结论:该法稳定、简便、重复性好,建立的氨基酸类成分指纹图谱特征性强,可为西洋参石斛药膳的质量评价和工艺优选提供参考。

Abstract:Objective:To establish the HPLC fingerprint of amino acida compounds from Panacis Quinquefolii Radix and Dendrobii Caulis in medicated diet. Methods:To apply 2,4qnitrofluorobenzene pre-column derivatization HPLC method. Separation was per- fomed on Kromasil C18 (250mm × 4. 6mm, 5μm) analytical column with mobile phase consisting of N, N-dimethylformamide- 0. 025mol . L^-1 sodium acetate ( 1 : 100,with 36% acetic acid adjust to pH6.0) and acetonitrile with the flow rate of 1.0mL .min^-1 and the column temperature at 40℃. The UV detection wave length was set at 360nm, the injection volume was 10μL and 10 batches were determined. Results:The fingerprint of Panacis Quinquefolii Radix and Dendrobii Caulis in medicated diet was established with 23 common peaks, 17 kinds of amino acid compsdition were identified, The similarity of 10 batches of samples was above 0.92. Conclusion: Each ingredient's amino acids showed in the characteristic fingerprint. The characteristic fingerprint of amino acid components can be used for the quality control.
