β细胞素基因多态性与2型糖尿病患者血脂异常相关性研究

摘    要:目的:探讨β细胞素基因外显子1TGC19GGC、5′非翻译区A-226G两个位点基因多态性与2型糖尿病患者血脂代谢的相关关系。方法:对130例2型糖尿病患者和96例健康对照者分别采用RFLP-PCR、SASP-PCR对两个位点进行基因多态性筛查。结果:①外显子1TGC19GGC研究中,2型糖尿病组总胆固醇、高密度脂蛋白、低密度脂蛋白、甘油三酯在两种基因型之间无显著性差异(P>0.05);健康对照组总胆固醇、甘油三酯、低密度脂蛋白在两种基因型之间无显著性差异(P>0.05),高密度脂蛋白在两种基因型之间虽无显著性差异(P>0.05),但是在TG基因型中有降低的趋势。②5′非翻译区A-226G研究中,2型糖尿病组总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白在两种基因型之间无显著性差异(P>0.05);对照组总胆固醇、高密度脂蛋白、低密度脂蛋白两种基因型间无显著性差异(P>0.05),甘油三酯在GG基因型中明显升高(P<0.05)。结论:①BTC基因外显子1TGC19GGC的研究中发现,在2型糖尿病组中该位点基因突变与血脂水平无明显相关性,在健康对照组中TG基因型与高密度脂蛋白水平降低有一定的相关性。②BTC基因5′非翻译区A-226G的研究中发现,在2型糖尿病组中该位点基因突变与血脂水平无明显相关性,在健康对照组中GG基因型与甘油三酯水平升高有一定相关性。

The correlation between betacellulin gene polymorphism and dyslipidemia in type 2 diabetes mellitus

Abstract:Objective:To screen single nucleotide polymorphyisms of BTC exon 1 TGC19GGC and 5rUTR A 226G and to analyze the correlation between BTC gene SNPs and Dyslipidemia. Methods: All T2 DM patients were consisted of 130 persons,Normal control group were consisted 96 persons ;Screened SNP of the exon 1 TGC19GGC by RFLP PCR,Screened SNP of the A-226G by SASP PCR. Results= @The exon 1 TGCIGGC mutation :TC,TG, LDL C, HDL-C were no significant differences between TG genotype and TT genotype in T2 DM group(P〉0.05) ; TC,TG,LDL-C were no significant differences between TG genotype and TT genotype in normal control group (P〉 0.05) ;The level of HDL c in blood was lower in TG genotype than in TT genotype in normal control group(P〉0. 05). @The 5' UTR A-226G mutation : TC, TG,LDL-C. HDL-C were no significant differences between two geno types in TeDM group(P〉0.05);TC.HDL-C,LDL-C were no significant differences between two genotypes in nor real control group (P〉0.05) ;The level of TG in blood was higher in GG genotype in normal control group(P〈0. 05). Conclusion: @In the study of BTC exon 1 TGCIGGC,the site SNP is not correlated with Dyslipidemia in T2 DM group ;the TG genotype is correlated with the level of HDL c in blood in normal control group. @In the study of BTC 5'UTR A-226G,the site SNP is not correlated with Dyslipidemia in T2DM group ;the GG genotype is signifi cantly correlated with the level of TG in blood in normal control group.
