小鼠牙胚fgf18的基因克隆和序列分析

摘    要:目的 从小鼠牙胚克隆成纤维细胞生长因子18(fgfl8)的全部编码区,测定并分析其基因序列。方法 用巢氏逆转录-聚合酶链反应(RT—PCR)扩增出fgfl8全部编码蛋白的基因,并克隆到PGEM—T载体,通过PCR及KpnⅠ和Hind Ⅲ限制性内切酶双酶切鉴定重组质粒,测定fgfl8序列。结果 测序结果表明从小鼠牙胚中成功扩增出fgfl8序列,经BLAST分析与已公布的小鼠fgfl8基因编码区完全一致。结论 fgfl8可能参与小鼠牙齿的发育和矿化。

Abstract:To clone the complete encoded sequence mouse fgf18 gene from mouse tooth germ,squence and analyze its sequence.Methods The complete encoded sequence mouse fgf18 gene has been amplified by nested RT- PCR from mouse tooth germ,the amplifid fragment was cloned into PGEM-Tvector and identified by means of PCR test and KpnI/HindIII digestion ,the cloned cDNA in this recombinant plasmid was sequenced.Results Full length cDNA of encoding mouse fgf18 gene has been cloned from mouse tooth germ and nucleotide homology analysis indicated the sequence was identical to that of gene bank.Conclusion fgf18 may have some effects on mouse tooth development and mineraliztion.
