白藜芦醇抑制人晶状体上皮细胞增殖的信号转导机制

摘    要:目的 探讨白藜芦醇(Res)对重组人碱性成纤维细胞生长因子(rhbFGF)诱导的人晶状体上皮细胞(HLEC)增殖的抑制作用,并从信号转导方面研究其抑制HLEC增殖的作用机制,为寻求防治后发性白内障的有效药物提供实验依据. 方法 采用rhbFGF诱导体外培养的HLEC增殖,流式细胞术(FCM)检测一定浓度的Res对增殖状态的HLEC增殖细胞核抗原(PCNA)蛋白表达的抑制作用:FCM检测不同的细胞内信号转导阻断剂(钙调素抑制剂W7、蛋白激酶C抑制剂H7)对其抑制作用的影响. 结果 Res对HLEC内PCNA蛋白表达有明显的下调作用(P<0.01);W7、H7均明显上调了HLEC内PcNA蛋白表达(P<0.01). 结论 Res有抑制HLEC增殖的作用,其机制可能是通过CaM、PKC等来介导的.
