葡萄糖转运蛋白1、3在非小细胞肺癌中的表达及其临床意义

摘    要:目的探讨葡萄糖转运蛋白1(Glucose transporter,Glut 1)和3(Glut 3)在肺癌及肺良性病变中的表达以及两者在肺癌发生发展过程中的关系.方法收集68例肺癌和17例肺良性病变组织,采用单克隆抗体Glut 1、Glut 3的免疫组化ABC法进行检测.结果68例肺癌组织中58例Glut 1阳性,表达率为85.3%;其中鳞癌100.0%表达,腺癌70.5%,Gluf 1在肺癌组织染色阳性率与分化程度、TNM分期及淋巴结转移等显著相关;Glut 3有18例在肺癌中染色阳性,表达率23.5%,其中鳞癌41.2%表达,腺癌11.8%,Glut 3在肺癌组织中表达与分化程度、TNM分期及淋巴结转移等无明显差异,Glut 3染色阳性对应的标本Glut1都染色阳性.在肺良性病变组织均未检测到Glut 1、Glut 3的表达.结论Glut 1在非小细胞肺癌中过度表达,且与肿瘤的分期、淋巴结转移、分化程度等密切相关,参与非小细胞肺癌的发生发展;Glut 3在少数非小细胞肺癌中存在过度表达,其表达发生在Glut 1过度表达之后.

Abstract:Objective To investigate the expressions of glucose transporter(GLUT) 1 and GLUT3 in lung cancer and benign pulmonary lesions and their relations during the occurrence and development of lung cancer. Methods Tissues of 68 non-small cell lung cancer(NSCLC) and 17 benign pulmonary lesions were collected,and then stained by monoclonal antibodies of GLUT1 andGLUT3 mediated ABc immunohistochemical method.Results The GLUT1 positive rate of lung cancer tissues was 85.3%(58 out of 68),which significantly correlated with cell differentiation degree,TNM stage and lymph node metastasis.Concretely,the GLUT1 positive rates of squamous cell carcinoma and adenocarcinoma were 100.0% and 75.5% respectively.18 of 68 lung cancer tissues were GLUT3 positive,and the positive rates of squamous cell carcinoma and adenocarcinoma were 41.2% and 11.8% respectively,which had no significant correlations with cell differentiation degree,TNM stage and lymph node metastasis.All GLUT3 positive samples were also GLUT1 positive.The expressions of GLUT1 and GLUT3 had not been detected in benign pulmonary lesions.Conclusions GLUT1 over-expression exists in NSCLC,which is closely related with TNM stages,lymph node metastasis and cell differentiation degree and involved in the occurrence and development of lung cancer.Over-expression of GLUT3 exists in a few of NSCLC,usually after the over-expression of GLUT1.
