PTEN和hMSH2在乳腺浸润性导管癌中的表达及临床意义

摘    要:目的 探讨抑癌基因PTEN和错配修复基因hMSH2在乳腺浸润性导管癌中的表达,以及与临床病理指标的关系。方法采用免疫组化EnVision法检测75例乳腺浸润性导管癌及30例正常乳腺组织中PTEN和hMSH2蛋白的表达情况。结果乳腺癌与正常乳腺组织相比,PTEN和hMSH2蛋白表达阳性率均明显降低,分别为64%和72%(P均〈0.05)。乳腺癌中PTEN表达缺失与组织分化差、腋窝淋巴结转移、临床分期有关,与患者年龄、肿瘤大小、C—erbB-2、ER、PR表达无关;hMSH2表达降低与肿瘤体积大、组织分化差、腋窝淋巴结转移有关,与患者年龄、临床分期、C—erbB-2、ER、PR表达无关;PTEN蛋白与hMSI-L2蛋白在乳腺癌中表达呈显著正相关(r=0.387,P〈0.01)。结论PTEN和hMSH2表达缺失与乳腺癌的发生发展及生物学行为密切相关;二者相互协同导致肿瘤形成和恶性程度增加。

Study on the expression of PTEN and hMSH2 in breast invasive ductal carcinoma and its clinical significance
Abstract:Objective To investigate the expression of the tumor suppressor gene PTEN and mismatch repair gene hMSH2 in breast invasive ductal carcinoma and their correlations with clinieopathologie parameters. Methods 75 cases of primary invasive breast carcinomas and 30 normal breast tissues were detected for FFEN and hMSH2 protein expression by means of immunohistochemical EnVision method. Results In breast cancers, the positive rate of PTEN and hMSH2 protein was 64% and 72% , respectively, significantly lower than that in the normal breast tissues (P 〈 0. 05 ). Reduced PTEN expression was significantly correlated with poor histological differentiation, axillary lymph node metastasis and late clinical stage( P 〈 0.05 ) , but not with age, tumor size, C - erbB - 2 expression, ER and PR status( P 〉 0. 05). Decreased expression of hMSH2 was correlated with tumor size, poor histological differentiation and axillary lymph node metastasis ( P 〈 0.05 ), but not with age, clinical stage, C - erbB - 2 expression, ER and PR status(P 〉 0. 05). There was a significant positive association between PTEN and hMSH2 expression( r = 0. 387 ,P 〈 0. 01 ). Conclusion Defective expression of PTEN and hMSH2 might play a cooperated role during multi - step breast carcinogenesis, and it has a closely relationship to the biological behavior of the cancer.
