薯蓣皂苷含药血清对乳鼠心肌细胞过氧化损伤的保护作用

摘    要:目的:采用血清药理学方法研究薯蓣皂苷对过氧化氢(H2O2)致心肌细胞过氧化损伤的保护作用。方法:采用血清药理学方法制备薯蓣皂苷含药血清(600 mg·kg-1)。体外培养原代乳鼠心肌细胞,随机分为DMEM对照组、过氧化损伤模型组、正常血清对照组、薯蓣皂苷低、中、高剂量干预组,分别加入不同处理因素干预2 h。除DMEM对照组外,其余细胞均以终浓度为100μmol·L-1的H2O2进行损伤2 h,MTT法测定细胞存活率,并检测各组心肌细胞培养上清液中乳酸脱氢酶(LDH)、丙二醛(MDA)的含量及总超氧化物歧化酶(T-SOD)及过氧化氢酶(CAT)的活性。结果:与DMEM对照组相比,损伤模型组细胞经100μmol·L-1 H2O2处理2 h后,活力降至(33.2±5.0)%(P<0.01);与损伤模型组及正常血清组比较,薯蓣皂苷低、中、高剂量干预组的心肌细胞存活率分别升至(42.3±2.5)%,(62.8±3.2)%,(74.3±2.8)%(P<0.01),呈现一定的剂量依赖性;与DMEM对照组相比,H2O2损伤后MDA、LDH含量明显升高,T-SOD、CAT活性降低(P<0.01),而薯蓣皂苷含药血清可剂量依赖性的降低损伤细胞MDA、LDH含量(P<0.01),升高CAT、T-SOD活性(P<0.01)。结论:薯蓣皂苷对H2O2诱导的心肌细胞损伤具有保护作用,机制与增强细胞抗氧化作用、减轻自由基和脂质过氧化物导致的细胞膜损伤有关。

Abstract:OBJECTIVE To study the effects and mechanism of dioscin on hydrogen peroxide(H2O2) injured cardiomyocytes with a serum pharmacological method.METHODS Prepared the serum containing dioscin(600 mg·kg-1) with serum pharmacological method.Cardiomyocytes derived from neonatal rats were cultured in Dulbecco Modified Eagle Medium(DMEM).The cardiomyocytes were injured with hydrogen peroxide(H2O2),and treated with the serum containing different concerntrations of dioscin.The survival rates of cardiomyocytes were measured with MTT method.The contents of malondialdehyde(MDA) and lactate dehydrogenase(LDH),and the activities of total superoxide dismutase(T-SOD) and catalase(CAT) were measured.RESULTS The survival rates of cardiomyocytes injured with H2O2 for 2 h were decreased significantly compared with normal cells(P<0.01).The content of MDA and LDH were increased significantly,but the activity of T-SOD and CAT were decreased.After treated with the different concentrations of drug-containing serum,the survival rates were increased with a dose-dependant manner(P<0.01).The contents of MDA and LDH were decreased and the activities of SOD and CAT were significantly increased(P<0.01).CONCLUSION Dioscin can protect the H2O2 injured cardiomyocytes by increasing antioxidation capability.
