降脂平肝汤对非酒精性脂肪性肝病大鼠肠源性内毒素血症的影响

摘    要:目的:观察复方中药降脂平肝汤对非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)大鼠肠源性内毒素血症(intestinal endotoxemia,IETM)的影响,探讨其作用机制。方法:21只Wistar大鼠随机分为正常组、模型组和中药治疗组。除正常组大鼠饲以普通饲料外,其余两组采用富含高不饱和脂肪酸的高脂饮食16周复制大鼠NAFLD模型,中药治疗组同时以复方中药降脂平肝汤混悬液灌胃。观察动物一般情况,计算肝指数、肝组织细胞脂变及炎症活动程度,检测肝组织Tch、TG、血清ALT、血浆肿瘤坏死因子α(Tumor Necrosis Factor-α,TNFα)和内毒素(Endotoxin,ET)含量等指标。结果:降脂平肝汤明显改善了NAFLD大鼠的肝指数、肝组织脂肪变程度、炎症活动程度,中药治疗组大鼠肝组织Tch与TG含量、血浆ALT活性与TNFα、ET含量均比模型组显著降低(P〈0.05,P〈0.01)。结论:降脂平肝汤减轻IETM,调节脂质代谢是其防治NAFLD的重要机制。

Effect of Jiangzhi Pinggan decoction on intestinal endotoxemia of fatty liver rats

Abstract:Objective: To study the effect of Jiangzhi Pinggan decoction on intestinal endotoxemia (IETM) of nonalcoholic fatty liver disease ( NAFLD ) rats. Methods: Twenty-one Wistar rats were randomly divided into the normal control group, model group and herbal therapy group. Except normal control group, the other two groups were fed with high-fat diet riched in unsaturated fatty acid, and the therapy group rats were treated with Jiangzhi Pinggan decoction. After 16 weeks, the TCh and TG in liver homogenatethe and serum ALT, ET, TNFα were measured. The liver histological and the ultrastructural changes were observed by light and electron microscopy. Results: Jiangzhi Pinggan decoction can evidently improve the liver pathology, decrease liver index, liver lipid and content of ALT, TNFα and ET ( P 〈 0. 05, P 〈 0. 01 ) . Conclusion: Jiangzhi Pinggan decoction can prevent and cure NAFLD by reduced IETM and adjust lipid metabolism.
