摘    要:目的 :探讨石蜡包埋组织切片的原位核酸杂交方法 ,以检测克山病 ( KSD)尸检心肌中的RNA。方法 :应用生物素标记的人工合成寡核苷酸探针 ,对 72例心肌石蜡包埋组织切片进行原位核酸杂交。结果 :各型克山病心肌石蜡组织切片的阳性杂交信号为 80 %~ 87.9% ,而正常对照仅为 14.3%。结论 :应用人工合成寡核苷酸之原位核酸杂交技术 ,对长期保存的石蜡组织进行回顾性研究是可行的。

Abstract:Objective: To explore the method of hybridization in situ in paraffin embedded myocardial specimens in order to examine RNA of Coxsackie virus of myocardial tissue from patients with Keshan disease(KSD). Methods: Hybridization in situ was performed by the use of the oligonucleotide probe by artificial synthesis labelled with biotin.Results:Positive hybrid signal was found in paraffin embedded myocardial specimens. The positive rates in acute,subacute and chronic KSD were 83%(10/12),87.9%(29/33) and 80%(16/20) respectively, but only 14.3%(1/7) in control.Conclusion: It is feasible to perform the reviewable study on paraffin embedded tissue kept for long term using hybridization in situ technology. [
