uPA在肝癌细胞株及肝癌组织中的表达及意义

目的:从mRNA水平探讨uPA在不同转移潜能肝癌及恶性黑色素瘤细胞株中的表达,从蛋白水平探讨uPA在肝癌组织中的代表表达及其与肝癌转移的关系。方法:检测uPA在不同转移潜能人肝癌及恶性黑色素瘤细胞株中的mRNA表达,用免疫组织化学方法检测uPA在肝癌组织中的蛋白表达。结果:uPAmRNA在高转移潜能肝癌和恶性黑色素瘤细胞株呈高表达,在相应低潜能转移细胞株呈低表达;uPA蛋白在肝癌组织中的表达明显高于癌旁组织和正常对照组,在有转移及静脉癌栓形成的肝癌组织阳性表达高于无转移及无癌栓者,癌巢边缘的癌细胞表达强于癌巢中央,血管内癌栓呈强阳性表达。结论:uPA在肝癌的的生长、侵袭和转移中起重要作用,可能作为治疗的操作、预防转移复发的对象。

The expression and significance of u-PA in human hepatocellular carcinoma cell lines and cancer tissues

Objective:T1o study the mRNA expression of uPA in human hepatocellular carcinoma and melanoma cell lines with different metastatic potential and the protein expression of uPA in hepatocellular carcinoma tissues and its relations with metastasis . Methods Northern blot was used to detect the mRNA expression of u-PA in hepatocellular carcinoma and melanoma cell lines with different metastatic potential; Immunohistochemistry was used to detect the protein expression of u-PA in hepatocellular carcinoma tissues. Results uPA mRNA show high and low expression in cell lines with high and low invasive potential respectively ; The positive expression of uPA protein in cancer tissues is significantly higher than in para-cancer tissues and in normal tissues,and the positive expression in cancer with metastasis and cancer embolus is higher than in that without metastasis and cancer embolus; The positive expression in the edged cells of the cancer mass is stronger than that in the center, all of the cancer embolus cells show strong positive stain. Conclusion uPA played an important role in the growth、invasion and metastasis of hepatocellular carcinoma, it can be used as marker of metastasis and prognosis of hepatocellular carcinoma in clinical practice,and may become the target for therapy .
