荷瘤鼠连续传代过程中肿瘤转移能力的演变及机制

摘    要:目的观察荷Lewis肺癌(LLC)的C57BL/6小鼠连续传代过程中肿瘤转移能力的演变及瘤组织中炎性因素的变化,探讨肿瘤在活体内的转移能力、演变及其与炎症的关系。方法LLC荷瘤小鼠连续传代,于第2、8、15代各传15只小鼠为早期组、中期组、晚期组,观察三组小鼠的肺转移情况,检测各组小鼠肿瘤组织中的微血管密度(MVD)及巨噬细胞、基质来源趋化因子-1(SDF-1)的表达情况。结果随着荷瘤鼠传代次数的增加,肿瘤的肺转移能力明显增强,肿瘤组织中巨噬细胞明显增多(P〈0.05);肿瘤组织中的MVD、SDF-1的表达明显增加,且二者表达的增强与巨噬细胞的增多呈正相关(P〈0.05)。结论Lewis肺癌在小鼠体内连续传代过程中其转移能力逐渐增强,且这种演变与肿瘤的炎症浸润有重要的关系

Abstract:Objective To observe the changes of neoplastic metastasis and the variation of inflammation in the serial passage of the bearing tumor mice and to explore the correlation between inflammation and neoplastic metastasis. Methods The bearing tumor mice was serial passaged to observe lung metastasis in the second, the eighth and the fifteenth generation, immunohistochemistry was used to detect the density of CD68 and the expression of CD31 and SDF - 1 on each group. Resluts With the numbers of passage increasing, neoplastic metastasis was gradually enhanced. The density of macrophage and the expression of CD31 and SDF - 1 gradually increase and the expression of CD31 and SDF - 1 were positively correlated to the density of maerophage ( P 〈 0.05). Conclusions The LLC' s ability of metastasis is gradually enhanced in the process of serial passage in mice and there is important connection between this dynamic variation and inflammation.
