实验性肝纤维化大鼠基质金属蛋白酶-2和组织型金属蛋白酶抑制剂-1mRNA表达及姜黄素的作用

摘    要:目的探讨大鼠肝纤维化过程中基质金属蛋白酶-2(MMP-2)、组织型金属蛋白酶抑制剂-1(TIMP-1)mRNA表达及姜黄素汤对其的影响。方法复制大鼠四氯化碳(CCl4)肝纤维化模型,分别以高、中、低剂量姜黄素处理,检测血清透明质酸(HA)、层粘连蛋白(LN)、三型前胶原(PCⅢ)和四型胶原(IVC)水平,检测肝组织纤维化程度和MMP-2及其抑制因子TIMP-1mRNA表达。结果CCl4成功诱导肝纤维化大鼠模型,姜黄素治疗能够降低血清肝纤维化指标及肝组织纤维化程度,姜黄素可以抑制CCl4诱导的MMP-2和TIMP-1mRNA表达,并能恢复MMP-2/TIMP-1的动态平衡。结论姜黄素可以通过减少MMP-2及其TIMP-1表达抑制肝纤维化。

Abstract:Objective To investigate MMP -2 and TIMP - 1 mRNA and the effect of curcumin in liver fibrosis rat. Methods Models of liver fibrosis in rats induced by subcutaneously injection of CCl4 were treated with curcumin ormalotilate as positive control. The levels of HA, LN, PC m and IVC in serum and histological changes in liver tissue were determined to reveal the event liver damage. MMP -2 and TIMP - 1 mRNA expression in liver tissues were determined by RT-PCR. Results Curcumin significantly reduced the levels of HA, LN, PC m and IVC in serum and liver fibrosis was also improved. Curcumin also inhibited MMP -2 and TIMP - 1 mRNA expression in live tissues induced by CCl4. Conclusions Curcumin may have therapeutic effect on liver fibrosis by inhibiting MMP -2 and TIMP - 1 expression.
