骨髓基质细胞对人多发性骨髓瘤细胞系自分泌白细胞介素6诱导作用的研究

摘    要:研究了小鼠骨髓基质细胞对诱导人白细胞介素6(IL-6)依赖性多发性骨髓瘤(MM)细胞自分泌IL-6的作用。结果表明:人MM细胞系XG-1培养上清(SN_1)没有可检测水平IL-6活性;小鼠骨髓基质细胞系MF-1培养上清(SN_2)仅有低水平IL-6活性(100U/ml);XG-1细胞和MF-L细胞/上清共同培养上清(SN_3,SN_4)均含极高的IL-6活性(1050U/ml),并可被IL-6单抗BE-8特异性中和,而且不同浓度的SN_3,SN_4可以促进其他依赖外源性IL-6的人MM细胞系(XG-2,4,6)生长。鉴此证实了小鼠骨髓微环境基质细胞可以诱导MM细胞自分泌IL-6,后者可促进MM细胞生长。

Abstract:he roles of the murine stromal cells in inducing anautocrine IL-6 loop in human IL-6-dependent myelomacell line (XG-1) were studied. The results showed thatno IL-6 activity in the cultured supernatant of XG-1 cells(SN_1) and very low IL-6 activity in the cultured super-natant of murine stromal cell line MF-L(SN_2) were de-tected, very high level of IL-6 in the coculturedsuprnatant (SN_3 , SN_4) of MF-L cells/supernatant andXG-1 cells was found. Meanwhile, the activity of IL-6from SN_3/SN_4, which can stimulate the growth of otherIL-6-dependent multiple myeloma (MM) cell lines (XG-2 ,4 , 6) , can be totally inhibited by the specific mono-clonal antibody to IL-6 (BE-8).The study indicated that the stromal cells from murinebone marrow could rapidly induce an autocrine IL-6 loopin human IL-6-dependent MM cells and this loop pro-mote the tumor cells'growth without any exogenous IL-6.
