葛根素对人胚肺成纤维细胞增殖及细胞外基质合成的影响

摘    要:目的观察葛根素对体外培养人胚肺成纤维细胞增生和凋亡以及细胞外基质形成的影响。方法用0~400gg/mL浓度的葛根素作用于体外培养的人胚肺成纤维细胞,24~96h后观察不同浓度的葛根素对成纤维细胞增殖活性及其功能的影响。MTT法观察成纤维细胞的生长活性,天狼猩红染色观察胶原生成情况。流式细胞仪测定细胞周期时相变化。结果80~400gg/mL浓度作用下24~72h范围内,葛根素可剂量和时间依赖性地抑制人胚肺成纤维细胞的增生(P〈0.05)。流式细胞仪检测结果发现,随着浓度的增大,G0/G1期细胞百分比逐渐上升,S期细胞百分比逐渐下降,说明细胞阻滞于G0/G1期。结论葛根素可在一定程度上抑制人胚肺成纤维细胞的增生,并诱导其凋亡,降低胶原的合成,作用呈时间和剂量依赖性。

Abstract:Objective To investigate the effect of puerarin on proliferation of human embro fibroblast and extraceUuar matrix. Method Human embro fibroblasts were incubated with 0-400 μg/mL puerarin for 24-96 h. The MTT method was used to assay the biological activities and extraceUuar matrix formation in different time and different dose of pueratin. The cell cycle distribution was detected by flow cytometric analysis. Result Incubation cells in presence of puerarin of 40-400 μg/mL for 24-72 h could significant inhibit cell proliferation and restrain the collagen synthesis in a dose- and time-dependent manner (P〈0.05). After treated with puerarin at different concentrations for 24 h, the cell number in G0/G1 phase was increased while in S phase decreased (P〈0.05). Cells were arrested in G0/G1 phase. Conclusion Puerarin significantly inhibits proliferation of human embro fibroblast, makes it arrest in G0/G1 phase and restrains the collagen synthesis, which is dose- and time-dependent in vitro.
