运动、EGCG和肉碱对肥胖大鼠体重、内脏脂肪及肝脏CPT1表达的影响

摘    要:目的:探讨运动、表没食子儿茶素没食子酸酯(EGCG)和肉碱(LC)对肥胖大鼠体重、内脏脂肪及肝脏肉毒碱棕榈酰转移酶1(CPT1)基因、蛋白表达的影响。方法:选取造模成功的肥胖大鼠50只,随机分为5组:肥胖对照组(F),运动组(S),运动+EGCG组(S+E),运动+肉碱组(S+L),运动+EGCG+肉碱组(S+E+L),每组10只。用蒸馏水将EGCG、LC溶解成无色透明的溶液,EGCG和LC的补充量均按80 mg/kg体重计算,灌胃容量为1 ml/100g体重(体重超过500g的大鼠,灌胃容量都按5ml配比),F组、S组大鼠则按照1 ml/100g体重灌胃蒸馏水。除F组外,其他四组大鼠进行低强度有氧跑台训练,负荷设定为16m/min,60min/day,大约相当于大鼠58%最大摄氧量。7周后比较各组大鼠体重、内脏脂肪系数、肝脏甘油三酯(TG)水平及肝脏CPT1基因及蛋白表达水平。结果:S组、S+E组、S+L组、S+E+L组大鼠体重显著低于F组(P<0.05),S+E+L组体重显著低于S组(P<0.05)。S+E+L组内脏脂肪系数显著低于F组(P<0.05)。S+L组和S+E+L组肝脏TG水平显著低于F组、S组和S+E组(P<0.05)。S组大鼠肝脏CPT1基因及蛋白水平与F组无显著差异(P>0.05),S+E组、S+L组、S+E+L组大鼠CPT1基因水平较F组和S组显著增高(P<0.05),S+L组和S+E+L组CPT1蛋白量较F组显著增加(P<0.05)。结论:运动、EGCG、LC联合使用可起到减重、减少内脏脂肪和改善肝脏甘油三酯水平的作用,改变肝脏CPT1表达可能是其机制之一。

Abstract:Objective The effect of exercise,and administration of epigallo catechin gallate(EGCG)and L-carnitine administration on the body weight,visceral fat and carnitine palmitoyl transferase1(CPT1) expression in obese rats was studied.Methods Fifty obese rats were randomly and equally divided into the obesity group(group F),exercise group(group S),exercise + EGCG intervention group(group S+E),exercise + l-carnitine intervention group(group S+L) and exercise + EGCG + l-carnitine intervention group(group S+E+L).The gastric perfusion amount was 1ml solution of EGCG and LC per 100g body weight with a total amount of 80mg per kg body weight.Rats in groups F and S were given the same amount of distilled water.All rats,except in group F,underwent treadmill training at the speed of 16m/min(equivalent to 58% VO2max),60min/day.After 7 weeks,the body weight,visceral fat coefficient,liver triglyceride(TG) level,liver CPT1 mRNA and the CPT1 protein level of the five groups were compared.Results The body weight of the rats in groups S,S+E,S+L,and S+E+L were lower than in group F(P < 0.05),and in group S+E+L was significantly lower than in group S(P < 0.05).The visceral fat coefficient in group S+E+L was significantly lower than in group F(P < 0.05).The liver TG levels of groups S+L and S+E+L were significantly lower than of group F,S,and S+E.There was no significant difference in CPT1 mRNA and CPT1 protein expression between groups F and S(P > 0.05),however,the CPT1 mRNA in groups S+E,S+L,and S+E+L was higher than in groups F and group S.The CPT1 protein expression of groups S+L and S+E+L was higher than that of group F.Conclusion Exercise in combination with administration of EGCG and L-carnitine could reduce body weight and visceral fat,and improve liver TG level of the obese rats,probably relating to the changes in CPT1 expression.
