摘    要:目的 观察丙酮酸对急性阻塞性肠缺血再灌注(I/R)的影响。方法 50只Wistar大鼠(幼龄40只和成年10只)分为5组:手术对照组,缺血组,再灌注组,丙酮酸组,成年动物再灌注组。在60min缺血和120min再灌注后,进行生化和组织病理学检测。结果 缺血组血清谷丙转氨酶(ALT)值和肠粘膜损害较手术对照组明显升高,超氧化物歧化酶(SOD)活性无明显变化;

Effect of pyruvate in intestinal ischemia-reperfusion injury

Abstract:Objective To evaluate the effect of pyruvate on intestinal ischemia reperfusion injury using immature rat model. Methods Fifty Wistar rats (40 immature rats and 10 adult rats) were divided into 5 groups: 1)control group (sham operated, n=10), 2) ischemia group (60 minutes of superior mesenteric artery occlusion, n=10), 3) re perfusion group (60 minutes of superior mesenteric artery occlusion followed by 2 hours of reperfusion, n=10), 4) pyruvate group (injection of pyruvate after 30 minutes of ischemia, n=10) and 5) adult reperfusion group (60 minutes of superior mesenteric artery occlusion followed by 2 hours of reperfusion). Serum levels of SOD and ALT were measured at the end of the experiment. Histological studies of intestine, liver and lung were also carried out.Results The plasma ALT level and mucosal injury score were significantly higher in the ischemia group than the control group ( P < 0.01 ) whereas the SOD activities were significantly decreased compared to the control group ( P < 0.01 ). The plasma ALT levels and mucosal injury scores in the pyruvate group were significantly lower in the pyruvate group compared to the reperfusion group ( P < 0.01 ).Conclusions Intestinal ischemia can damage the intestinal mucosa and liver function in immature rats. Reperfusion can lead to further injury. Pyruvate administered before reperfusion may attenuate but not completely block the liver and mucosa damage. The ischemia reperfusion causes more damage to the intestine and liver in immature rats compared to the adult rats.
