摘    要:目的建立高效液相色谱法测定十味降糖泡腾颗粒中葛根素含量的方法。方法采用Hypersil C18色谱柱(250.0 mm×4.6 mm,5.0μm),甲醇-0.025 mol.L-1磷酸盐(pH 3.5)(14)为流动相,检测波长为250 nm,流速1.0 mL.min-1。结果理论塔板数按葛根素峰计算为7 600,供试品中葛根素能与其他杂质峰很好的分离,分离度R>1.5,峰形对称,拖尾因子为1。葛根素2.515~100.600 mg.L-1呈良好的线性关系(r=0.999 9),平均加样回收率和相对标准偏差分别为98.58%和2.40%。结论本法准确可靠,可用于测定十味降糖泡腾颗粒中葛根素的含量。

Abstract:Objective To establish a quantitative analysis method of puerarin in Shiwei Jiangtang effervescent granules by high performance liquid chromatography(HPLC).Methods HPLC with Hypersil C18 column(250.0 mm×4.6 mm,5.0 μm) was used,methanol-0.025 mol·L-1 phosphate(pH 3.5)(14) as the mobile phase,at the detection wavelength of 250 nm and the flow rate of 1.0 mL·min^-1.Results The number of theoretical plates of the column was 7 600,calculated with reference to the peak of puerarin.The puerarin of test sample was well separated from other impurities with separating degree R1.5,trailing factor was 1.The calibration curve of puerarin was a good linear relationship in the range of 2.515-100.600 mg·L^-1(r=0.999 9).The average recovery rate was 98.58%,and rlative standard deviation was 2.40%.Conclusion This method is accurate,reliable and can be used for the determination puerarin in Shiwei Jiangtang effervescent granules.
